First. Ask yourself what you want to achieve.
Secondly. What are the real benefits of Flash you can't do with other techniques.
Trend is to use AJAX within pages to load/scroll through certain parts, transitional effects, overlays etc etc. That way you can still be accessible to search engines and for users to use the back button as a navigational tool.
Unless you're into information graphics, advanced storytelling. Go with regular HTML/CSS. Animation, just for the sake of it can be tiresome after a few visits.
Most good pages these days are built strictly with CSS/Javascript and that gives you a lot of freedom for what you want.
That being said. Any serious page should have a fallback solution for those without Javascript turned on. It's a bit more common than you might think.
But I love the good stuff as much as anyone else. Here are some great examples of how to use Javascript/AJAX
